Entre sus obras de juventud se encuentran Der Logische Aufbau der Welt (La estructura lógica del mundo) (1928) y Scheinprobleme in der Philosophie (Pseudoproblemas de filosofía) (1928). En 1931 fue invitado como profesor a la Universidad de Praga, donde permaneció hasta 1934 y publicó su libro Logische Syntax der Sprache (Sintaxis lógica del lenguaje) (1934). Sus convicciones humanistas y socialistas lo llevaron a huir del régimen nazi en 1935. Emigró a Estados Unidos, en donde fue recibido por la Universidad de Chicago. Allí profundizó sus investigaciones de semántica formal y en 1942 publica su Introduction to Semantics (Introducción a la semántica); en 1947 la primera edición de Meaning and Necessity. A Study in Semantics and Modal Logic (Significado y necesidad. Un estudio de semántica y lógica modal). Más tarde, formó parte del Instituto de Estudios Avanzados en Princeton, y en 1954 fue invitado por UCLA, en donde enseñó filosofía hasta el final de su vida. Una segunda edición revisada y ampliada de Significado y necesidad, fue publicada por la Universidad de Chicago en 1956. Este libro constituye una valiosa exposición de una teoría del significado que surge a partir del trabajo de Gottlob Frege, y que ha tenido una enorme influencia en el campo de la semántica. Se trata de un libro sistemático que intenta reunir de manera metódica un cúmulo de cuestiones que muchas veces encontramos dispersas en la obra de otros autores.
